Abstract:

            The purposes of this research are to study the breaking of seed dormancy and microrhizome induction of Hedychium spicatum G. Lodd., seed dormancy breaking could induce the germination of seeds. The highest percentage of seed germination was 87.50% with germination time at 18 days when seeds were soaked in water for 24 hrs and seed coats were scarified. For microrhizome induction, microrhizomes were induced at thebase of the in vitro derived shoots upon transfer to Murashige and Skoog medium (MS) containing various concentrations of sucrose (30, 60, 90 and 120 g/L) with or without 5 g/Lactivated charcoal for 16 weeks. It was found that explants cultured on the medium with or without activated charcoal found microrhizome formation. The explants cultured on the medium with activated charcoal found the number of microrhizomes higher than explants cultured on the medium without activated charcoal. The maximum number of microrhizomes was in the medium containing 60 g/Lof sucrose with 5 g/L activated charcoal (3.50 ± 0.54 microrhizomes).
