Marcania grandiflora Imlay or ‘Gong Gang Nam Juad’ is an endemic plant found in Kanchanaburi and adjacent provinces of Thailand. M. grandiflora belongs to Acanthaceae family and is also the only member in genus Marcania. To obtain better understanding and useful information for conservation of this species, microsatellite or SSR (Simple Sequence Repeat) markers were developed specific to this plant. To develop the new markers, microsatellite enriched library of M. grandiflora was constructed using AG and CAG oligonucleotide probes to enrich the microsatellite fragments, then the microsatellites were identified in order to design the microsatellite primers. Of 88 sequences from the M. grandiflora library, 63 (71.6%) of microsatellite motifs were found. The abundance motifs in the library were AG/TC and AGC/TCG that found 29 and 12 sequences, respectively. In total, 63 microsatellite primers were design. These SSR primers will be used to evaluate the genetic diversity among the M. grandiflora populations found in Kanchanaburi and adjacent areas. The results of this project will be useful for establishing conservation plan of this endemic plant in the future.
