Banana (MusaL.) has recently been classified as one of the foods causing allergic reactions. Anumber of patients with allergy to banana have been reported in Thaipopulation, but therelevant allergens have not been described so far. To date, five proteins havebeen characterizedas banana allergens. Among them, beta-1,3-glucanase, is identified as a potent cross-reactiveallergen among rubber tree latex, pollens and some fruits. Thus, this study aimed to cloneand characterize beta-1,3-glucanase cDNAs from four banana cultivars: ‘Kluai Hom Thong’, ‘KluaiKhai’, ‘Kluai Namwa’ and ‘Kluai Lep Mue Nang’. The cDNAs encoding beta-1,3-glucanasefrom four banana cultivars were successfully cloned. All cDNAs contained an open readingframe of 1,023 bp encoding a protein of 340 amino acids with the predicted molecular massof 36.4 kDa and pI value of 8.83. Sequence analysis demonstrated that the deduced aminoacids from four banana cultivars shared 97–99% identity, suggesting their potential forallergenic cross-reactivity. This is the first report on molecular cloning ofcDNA encoding anallergenic protein from banana cultivars in Thailand. However, theallergenicity ofbeta-1, 3-glucanase among Thai patients suffering from banana allergy should befurther investigated.
